Localized Oxygen Exchange Platform for Intravital Video Microscopy Investigations of Microvascular Oxygen Regulation

Intravital microscopy has proven to be a powerful tool for studying microvascular physiology. In this study, we propose gas exchange system compatible with intravital that can used impose perturbations small localized regions in skeletal muscles or other tissues imaged using conventional inverted microscopes. We demonstrated the effectiveness of by locally manipulating oxygen concentrations rat extensor digitorum longus muscle and measuring resulting vascular responses. A computational model transport was partially validate localization changes tissue, saturation red blood cells flowing through capillaries were measured as surrogate local tissue oxygenation. Overall, have approach study dynamic spatial responses challenges microenvironment muscle.
